The dietary supplement industry thrives on a simple promise: enhance performance, slow aging, optimize health. Yet behind the marketing and often exaggerated claims, a small number of molecules stand out due to an unusually dense volume of research and a quality of evidence rarely matched in the nutritional field. Among thousands of products available, five substances consistently emerge in scientific literature: creatine monohydrate, caffeine, concentrated proteins, beta-alanine, and vitamin D.

Creatine: The Most Robust Supplement for Performance

Creatine holds a unique position. With over a thousand studies published since the 1990s, it is the most researched supplement in the realm of physical performance. Meta-analyses converge: creatine supplementation significantly increases maximal strength, notably measured by one-repetition maximum, and promotes lean mass accumulation.

The mechanism is well described. By increasing muscle phosphocreatine stores, creatine enhances the rapid resynthesis capacity of ATP during intense, short bursts of effort. It thus acts on the phosphagen energy system, central to explosive contractions.

More recently, research has expanded into the brain. Creatine appears to improve working memory and reduce mental fatigue, especially in situations of sleep deprivation or cognitive stress. Available data suggest a daily dose of three to five grams is sufficient to saturate muscle stores, with a favorable long-term safety profile in healthy individuals.

Caffeine: An Ancient Molecule with Modern Evidence

Caffeine, an ancient and ubiquitous molecule, also benefits from a solid scientific corpus. Recent meta-analyses, particularly in team sports like football, confirm its ergogenic effect. It improves agility, power, and endurance while reducing perceived exertion.

Its primary mechanism relies on antagonizing adenosine receptors in the central nervous system, which increases alertness, vigilance, and mobilization of energy substrates. Effectiveness appears optimal at doses between three and six milligrams per kilogram of body weight, administered about an hour before exercise.

Some studies also suggest a possible synergy with creatine, although exact interactions remain debated. However, caffeine is not without side effects, especially in sensitive individuals, with risks including anxiety, sleep disturbances, and increased heart rate.

Concentrated Proteins: Useful When Diet Falls Short

Protein powders, whether whey or collagen, form another cornerstone of sports supplementation. A recent network meta-analysis pooling several dozen studies confirms the benefit of additional protein intake to support hypertrophy and muscle recovery.

Whey, rich in leucine, effectively stimulates muscle protein synthesis via activation of the mTOR pathway. Collagen, long considered incomplete regarding essential amino acids, is experiencing renewed interest. Some recent data suggest notable effects on strength and lean mass, possibly through improved connective tissue integrity and muscle force transmission.

Interpretation of these results must remain cautious, as methodologies vary across studies. Timing of ingestion, particularly within the post-exercise window, seems to optimize the anabolic effect.

Beta-Alanine: The Buffer for Intense Efforts

Beta-alanine targets disciplines requiring intense efforts of medium duration, typically between one and four minutes. Its interest lies in its role in synthesizing muscle carnosine, a dipeptide that acts as an intracellular buffer against hydrogen ion accumulation.

By limiting muscle acidification, beta-alanine delays fatigue onset during prolonged anaerobic efforts. Meta-analyses confirm modest but significant gains in this type of exercise. Effective doses generally range from two to six grams per day, divided to minimize paresthesia, a frequently reported transient tingling sensation.

Vitamin D: Correcting Deficiency, Not Creating Miracles

Vitamin D occupies a different position. More than an ergogenic aid, it acts as a steroid hormone involved in regulating bone, immune, and muscle metabolism.

Studies show that correcting a deficiency improves muscle strength and reduces injury risk, especially in individuals living at high latitudes or with limited sun exposure. Recommended doses vary depending on initial status but often range between two thousand and four thousand international units per day.

Benefits appear most pronounced in deficient individuals, underscoring a fundamental principle: a supplement produces a measurable effect only if it corrects a deficiency or optimizes a limiting function.

Omega-3: Overall Health More Than Pure Performance

On the periphery of this restricted group lies another widely studied category for overall health: long-chain omega-3s, notably EPA and DHA. While their role in pure performance remains debated, abundant data exist regarding cardiovascular health, cognitive function, and inflammation modulation. Recent studies also suggest a favorable effect on muscle recovery and immune response to training.

Not All Supplements Are Equal

This overview reveals a frequently overlooked reality. Not all supplements are created equal, and the majority of commercial products lack a scientific foundation comparable to these few molecules. It also reminds us that effectiveness depends on context. Creatine supplementation will not have the same impact on a strength athlete as on a sedentary individual. Vitamin D correction will not transform an already optimal status.

The most rational approach remains evidence-based, prioritizing accordingly. Creatine likely offers the best balance between cost, safety, and demonstrated efficacy. Strategically used caffeine enhances acute performance. Proteins support muscle adaptation when diet is insufficient. Beta-alanine targets specific needs. Vitamin D corrects a common deficiency in modern populations.

Science does not validate the illusion of a shortcut to perfect performance or health. Rather, it shows that certain well-identified levers can amplify existing physiological mechanisms. The key is knowing why to activate them and for whom.
